The trail has been closed from mile marker 2.5 to marker 3.5, measured eastward from downtown.
The closure will last at least two weeks, according to the Port Angeles Parks Division.
The slide, which was reported to the city Wednesday morning, is larger and located slightly east of where another slide closed the trail last May.
That slide, between Morse Creek and the site of the former Rayonier pulp mill, sent earth more than 30 feet wide and 8 feet deep - plus two trees - across the popular running/cycling route.
It cost $20,000 to clear.
City parks personnel barricaded the slide area Wednesday.
The slide area is impassable and considered dangerous, said city spokeswoman Teresa Pierce.
People should stay clear of the site because other slides could occur due to recent heavy rains, she said.
The Olympic Discovery Trail, paved except for some of the Rayonier site, runs from Ediz Hook nearly to Sequim.
Eventually, it will connect LaPush on the Pacific Coast with Port Townsend.
